It may be just a wacky idea, but would it be possible to play the media in my home PC over the Internet? I mean, I know it can be accessed locally, but say I take my XBMC system to work, and would like to access the media from my home PC there. Would it be possible? What would I need?

you would need the media accessible via a protocol you can access over the net from your home to your work. Something like FTP or HTTP perhaps. in the case of http for example, you would need port 80 forwarded on home router to the webserver. (you can find guides on this stuff on the net)

Isn't plex essentially just a stripped down version of xbmc? Never used it so could be very wrong there.
Like buzz says you could do it via http, ftp etc. but as xphazer pointed out you'll need a good upload speed for that. Using addons to stream will most likely work better as they use dedicated fast servers and of course you'll have access to much more content.
